Moderna shares slide as investors lock in gains after historic cancer vaccine rally

Moderna (NASDAQ:MRNA) shares fell sharply in premarket trading on Thursday as investors took profits following the biggest one-day rally in the biotechnology company’s history, while concerns over valuation added to the selling pressure.

The stock dropped 13.0% to $151.78 before the opening bell, having closed the previous session at $174.38 following an extraordinary surge triggered by positive late-stage cancer vaccine results.

Phase 3 cancer vaccine success sparked record rally

Moderna shares more than doubled on August 19 after the company and Merck announced successful results from the Phase 3 INTerpath-001 study of their personalised mRNA cancer vaccine, intismeran autogene.

The trial, involving more than 1,100 patients with high-risk melanoma, met both its primary endpoint and key secondary endpoints.

The result represented an important milestone for the wider mRNA field, marking the first successful large-scale late-stage cancer trial involving an mRNA-based therapy and potentially strengthening Moderna’s prospects beyond its established vaccine business.

However, the scale and speed of Wednesday’s repricing left the shares vulnerable to investors locking in gains during the following session.

Analyst targets highlight valuation concerns

Fresh analyst commentary also contributed to caution around Moderna’s valuation after the stock’s dramatic advance.

RBC Capital raised its price target substantially to $130 from $45 but maintained a Sector Perform rating. Bank of America also upgraded Moderna and increased its target to $170.

Although both moves represented major improvements from previous expectations, the revised targets remained below Moderna’s Wednesday closing price of $174.38, reinforcing concerns that much of the potential value from the Phase 3 breakthrough had already been reflected in the shares.

Technical indicators provided another reason for caution. The stock became heavily overbought following its record rally, with the RSI moving to extreme levels.

Market analysts also noted that biotechnology stocks can frequently surrender 20% to 30% of their initial gains in the sessions following major binary clinical events as traders crystallise profits and investors reassess valuations.

Financial losses remain a consideration for investors

Despite the potentially transformative clinical development, Moderna’s underlying financial position continues to present challenges.

The company reported a net loss of $782 million for the second quarter of 2026 and continued to generate negative operating cash flow, leaving investors to balance the potential commercial value of its oncology pipeline against ongoing losses.

Those fundamentals have become more significant following the rapid increase in Moderna’s market valuation, particularly as investors assess how long it could take for the cancer vaccine programme to translate into meaningful commercial revenue.

Moderna decline remains company-specific

Broader equity markets offered little explanation for Thursday’s weakness, with the S&P 500, Nasdaq and Dow Jones all trading broadly flat.

That suggests Moderna’s premarket decline was predominantly driven by stock-specific factors rather than a wider deterioration in risk sentiment.

Other biotechnology companies benefited from Wednesday’s clinical breakthrough, including fellow mRNA oncology developer BioNTech, which recorded sympathy gains.

Thursday’s selling, however, was concentrated more heavily in Moderna as traders who entered during the previous session’s surge moved to secure profits.

The Phase 3 success remains a potentially significant long-term catalyst for Moderna, but the stock’s extraordinary move from below $65 to above $174 in a single session created an unusually demanding valuation backdrop. The subsequent pullback therefore reflects a period of consolidation as investors reassess how much of the cancer vaccine opportunity should already be priced into the shares.
