Cognition Therapeutics Secures US Patent for Zervimesine Through 2045 Ahead of Planned Phase 3 Development

By Fiona Craig | October 13, 2026, 6:59 AM

Cognition Therapeutics, Inc. (NASDAQ:CGTX) has received a new composition of matter patent from the US Patent and Trademark Office (USPTO) covering a crystalline form of its investigational drug candidate zervimesine (CT1812), extending intellectual property protection for the technology until 2045.

The patent, numbered 12,759,979, covers a specific polymorphic crystalline form of zervimesine, a proprietary manufacturing process and therapeutic methods associated with several neurological and ophthalmic conditions.

The protected applications include Alzheimer’s disease, dementia with Lewy bodies (DLB) and dry age-related macular degeneration (AMD).

Cognition Therapeutics said the newly patented form offers manufacturing and pharmaceutical advantages that could support larger-scale production as the company prepares for potential late-stage clinical development.

The patent has a natural expiration date in 2045 and could qualify for an extension of up to five additional years if the treatment receives approval from the US Food and Drug Administration (FDA), subject to applicable requirements.

New Patent Covers Drug Formulation and Manufacturing Process

The USPTO’s decision provides Cognition Therapeutics with intellectual property protection for a novel crystalline form of zervimesine.

The patent covers the drug’s polymorphic structure, which refers to a particular arrangement of molecules within a crystalline solid.

Different crystalline forms of the same compound can have distinct physical and chemical properties, potentially affecting stability, processing and manufacturing characteristics.

For pharmaceutical developers, these properties can be important when preparing a drug candidate for larger clinical trials and potential commercial production.

The patent also includes protection for the company’s proprietary production process and specified therapeutic uses.

By securing rights to the new crystalline form, Cognition Therapeutics has strengthened its intellectual property portfolio around zervimesine as it evaluates the drug across several indications.

However, patent protection does not establish the treatment’s clinical effectiveness or guarantee regulatory approval.

Improved Drug Properties Could Support Phase 3 Manufacturing

Cognition Therapeutics Chief Medical Officer Anthony Caggiano, M.D., Ph.D., highlighted the potential manufacturing benefits of the patented crystalline form.

According to Caggiano, the new version of zervimesine “has an improved pharmaceutical and physicochemical profile, making it better suited to the large-scale manufacturing needed for Phase 3 trials and commercial production.”

The company expects these characteristics to support the production requirements associated with later-stage clinical development.

Manufacturing consistency and scalability are important considerations for investigational treatments advancing towards registrational studies, which are generally designed to generate evidence for regulatory submissions.

Cognition Therapeutics plans to begin manufacturing the newly patented form of zervimesine during the first quarter of 2027.

The material is intended for use in a planned registrational clinical trial involving patients with dementia with Lewy bodies who experience psychosis.

However, the manufacturing timetable and clinical development plans remain dependent on the outcome of discussions with the FDA.

FDA Meeting Planned to Finalise DLB Trial Endpoints

Cognition Therapeutics has scheduled a meeting with the FDA to discuss and finalise the selection of clinical endpoints for its proposed registrational programme in dementia with Lewy bodies.

Clinical endpoints are the measures used to assess whether an investigational treatment provides a meaningful benefit to patients.

Their selection is particularly important in registrational trials because the resulting evidence may form part of a future application for regulatory approval.

The company intends to advance zervimesine into a study targeting patients with DLB who experience psychosis.

The development strategy is supported in part by findings from the Phase 2 SHIMMER study, which Cognition Therapeutics has said produced signals relevant to the proposed indication.

The company has not indicated that the FDA has approved the planned trial design or confirmed a definitive start date for the registrational study.

A favourable outcome from the upcoming regulatory meeting would support the company’s plans to begin manufacturing the new crystalline form in early 2027.

Zervimesine Under Evaluation for Alzheimer’s Disease

Zervimesine is an investigational oral treatment designed for once-daily administration.

The drug is currently being evaluated in the Phase 2 START study involving patients with mild cognitive impairment and early Alzheimer’s disease.

The study forms part of Cognition Therapeutics’ broader clinical development programme assessing the potential applications of zervimesine in neurological conditions.

The company has also completed Phase 2 studies involving patients with mild-to-moderate Alzheimer’s disease.

Alzheimer’s disease is a progressive neurological condition associated with cognitive decline, memory impairment and changes in everyday functioning.

Although zervimesine is being investigated as a potential treatment, its safety and effectiveness have not been established for regulatory approval.

Further clinical evidence will be required to determine whether the drug provides sufficient benefit in the patient populations being studied.

Previous Studies Include Dementia With Lewy Bodies and Dry AMD

Beyond Alzheimer’s disease, Cognition Therapeutics has completed Phase 2 studies of zervimesine in dementia with Lewy bodies and geographic atrophy associated with dry age-related macular degeneration.

DLB is a neurodegenerative condition that can affect cognition, movement, behaviour and perception.

The company’s planned registrational programme is focused specifically on patients with DLB who experience psychosis.

Cognition Therapeutics has identified findings from its Phase 2 SHIMMER study as part of the rationale for pursuing further development in this patient group.

The drug has also been investigated in geographic atrophy, an advanced form of dry AMD that causes progressive damage to the retina and can result in vision loss.

The newly granted patent includes therapeutic methods relevant to these indications, providing intellectual property coverage across several areas of the company’s research programme.

However, the existence of patent protection does not indicate that clinical development in each indication will necessarily proceed to commercialisation.

Patent Strengthens Long-Term Intellectual Property Position

The new patent provides Cognition Therapeutics with a potential intellectual property framework extending through 2045.

If zervimesine ultimately receives FDA approval and meets the relevant statutory criteria, the patent could be eligible for an extension of up to five years.

Any extension would depend on the applicable regulatory and patent requirements and should not be considered guaranteed.

For Cognition Therapeutics, the patent adds protection around a formulation intended to support the next stage of clinical development.

The company’s immediate priorities include finalising the proposed DLB registrational trial endpoints with the FDA and preparing for manufacturing activities planned for the first quarter of 2027.

The progression of zervimesine will depend on regulatory feedback, clinical trial outcomes and the company’s ability to execute its development programme.

The latest patent award represents an intellectual property milestone, while the drug remains investigational and has not received FDA approval for any indication.
